pASK-IBA4C is a classic cloning vector devised for E. coli expression of recombinant proteins with the Strep-tagII fused to the N-terminus. It carries the chloramphenicol resistance cassette for selection of transformed E. coli cells, F1 and ColE1 origin for a high plasmid copy number, a multiple cloning site with recognition sites for several common restriction enzymes (e.g. EcoRI, SmaI, BamHI, SalI, PstI), the inducible tetracycline promoter/operator for the regulated expression of proteins, and the ompA signal for periplasmic secretion of the recombinant protein.The vector can be combined with any E. coli strain since the tet-promoter works independently of the genetic background of E. coli. Expression of the recombinant protein is induced by addition of anhydrotetracycline, which is a derivative of tetracycline that does not exhibit antibiotic activity. During the secretion into the periplasmic space the ompA signal sequence will be cleaved off.

To construct Myr-HA-Akt1, the c-Src myristoylation sequence was fused in frame to the N terminus of the HA-Akt (wild-type) coding sequence. Myr-HA-Akt1 is a constitutively active mutant of Akt1, and it is also referred as Akt(ca).Akt1/PKB is activated in response to many extracellular signals and play key roles in regulating diverse cellular processes, including cell proliferation, differentiation, apoptosis, angiogenesis and glycogen synthesis etc. Akt1 is activated by the phosphorylation of the T loop of their kinase domain by PDK1 and by phosphorylation of a residue located C-terminal to the kinase domain in the hydrophobic motif. The Akt family of kinases includes Akt1, Akt2 and Akt3.

GCaMP is a genetically encoded calcium indicator (GECI) which was created from a fusion of green fluorescent protein (GFP), calmodulin, and M13, a peptide sequence from myosin light chain kinase. Aurora related kinase-1 (ARK-1, STK15, STK6, Aurora2) and -2 (ARK-2, STK12, Aurora1) are centrosome-associated serine/threonine kinases that regulate centrosome separation, bipolar spindle assembly and chromosome segregation during mitosis. First characterized in human colorectal cancers, ARK-1 and -2 are closely related to Drosophila aurora kinases.
